Full length PCR product was purified by electrophoresis in a 1% agarose gel using the QIAquick Gel Extraction kit (Qiagen) and was inserted into a pDONR/Zeo entry vector using a PCR Cloning System with the Gateway Technology kit (Invitrogen). Starting with this entry clone, the hASP DNA construct was cloned into a pDEST15 expression vector to obtain the DNA for the GST fusion protein. The expression vector was used to transform BL21 DE3 cells (Invitrogen). Transformants were selected on LB medium plates containing 50 μg ml−1 ampicillin, and the sequence was confirmed by DNA sequencing, which was carried out using the dideoxychain-termination procedure developed by Genome Express

est ce que vous connaissez un site qui résume les bases ..les ligations au iveau de plasmides etc ....
